Rachele ha 8 anni, vive in Umbria ed è una bambina solare, curiosa e piena di vita.
Fino a poco tempo fa la sua era una vita normale, fatta di scuola, giochi e sogni per il futuro. Nel corso del sesto anno di vita sono arrivati i primi segnali: difficoltà a leggere la lavagna, atassia e difficoltà nel fare le scale.
Gli esami effettuati nel 2024 hanno mostrato gravi alterazioni retiniche. L’elettroretinogramma e la Tomografia Ottica Computerizzata hanno evidenziato segni compatibili con una distrofia retinica maculare.
Inizialmente nel pannello genetico non erano state individuate mutazioni, ma a marzo 2025, grazie a un’intuizione del team medico che ha in carico Rachele, i dati sono stati rianalizzati includendo il gene CLN3.
Il 22 maggio 2025 è arrivata la conferma: delezione in omozigosi degli esoni 8 e 9 del gene CLN3, compatibile con la Ceroidolipofuscinosi neuronale di tipo 3 (Malattia di Batten), una patologia genetica ultra-rara e neurodegenerativa.
Questa malattia, in pochi anni, priva i bambini della vista, delle capacità motorie e cognitive, fino a compromettere la vita stessa.
Un’opportunità unica: la terapia genica negli Stati Uniti.
Grazie alla diagnosi precoce, Rachele è stata inserita in un trial clinico sperimentale di terapia genica negli USA, attualmente l’unica speranza concreta per rallentare la progressione della malattia.

Help Rachele Access Life-Saving Gene Therapy in the United States.
Rachele is an 8-year-old girl from Umbria, Italy. She is bright, curious, and full of life.
Until recently, her days were filled with school, playtime, and dreams for the future. But at age six, the first symptoms appeared: difficulty seeing the blackboard, problems with balance, and trouble climbing stairs.
In 2024, medical exams revealed severe retinal damage. Further testing, an electroretinogram and Optical Coherence Tomography, showed signs consistent with macular retinal dystrophy.
At first, no genetic mutations were identified. But in March 2025, thanks to the intuition of her medical team, the data was reanalyzed to include the CLN3 gene.
On May 22, 2025, the diagnosis was confirmed:
a homozygous deletion of exons 8 and 9 of the CLN3 gene, compatible with Neuronal Ceroid Lipofuscinosis type 3 (Batten Disease) an ultra-rare, neurodegenerative genetic disorder.
Batten disease progressively steals children’s vision, motor skills, and cognitive abilities, eventually threatening their lives.
But for Rachele, there is still hope.
