#FarmaCovid: un farmaco efficace contro il COVID19

Ci troviamo in una fase molto seria della pandemia da #Coronavirus. Di giorno in giorno il numero dei nuovi #contagi aumenta, e così la pressione sul sistema #sanitario. Ci attendono mesi difficili, si deciderà nei prossimi giorni, nelle prossime settimane, come sarà il nostro inverno, il nostro #Natale. Lo decideremo noi tutti, con le nostre azioni. Ora dobbiamo fare di tutto per fermare questo virus. Ogni giorno è prezioso. #Sicurezza, #distanziamento, protezione certo, ma anche il nostro Presidente della Repubblica #Mattarella ribadisce: “la #Ricerca è l’antidoto”. Un vaccino ci proteggerà dall'infezione, ma un #antivirale ci aiuterà a salvare chi è stato contagiato gravemente.
Per questo ogni sostegno alla ricerca di antivirali efficaci è adesso indispensabile.

#FarmaCovid continua la sua lotta contro il #virus. I fondi raccolti ad oggi hanno permesso di individuare un nuovo candidato #farmaco come potenziale anti-#COVID 19 (in fase di pubblicazione scientifica), e capace di inibire anche altri coronavirus, per cui utile per contrastare eventuali future #emergenze.

We are seeing the #Covid-19 pandemic growing rapidly again. The number of Covid-19 #cases increases by the day and so does the burden on the #healthcare system. The summer is over and now we need to face a challenging period. The next days or weeks will determine how we will spend our winter, our #Christmas. All this depends on us, on our actions. We need to do whatever it takes to defeat this virus. Each day is precious. #Security, #social distancing and protection are critical, but as the President of our Republic recently said “Research, that’s the antidote”. A vaccine will protect us from infection, but an antiviral drug will save those who have been already seriously infected. For this reason, the research on #antiviral drug must be supported, now.

FarmaCovid keeps on fighting the virus. The financial support we have received so far was critical, allowing for the identification of a new #candidate drug against COVID-19 (the scientific #publication will be soon available) and, also, #effective against other Coronaviruses.
We need drugs like these in order to face possible health emergencies of the #future.

The FarmaCovid Project

The funds raised will support the activities of the FarmaCovid project; in particular, Cnr-Ibf and ICGEB will be able to:

• Purchase an increasing number of drug candidates (15,000 euros) to be tested on viral proteins and directly on human cells infected with SARS-CoV-2 (25,000 euros); these activities already started on a small scale, but we need to increase the chances of success and accelerate the results of this first phase. Based on the funds raised, the researchers plan to complete this phase within the next six months.

• Select 2-3 best drug candidates, to study their activity over toxicity (20,000 euros for each selected drug); these tests are essential for subsequent studies with patients, and with adequate resources this phase could be completed within six months following the first phase.

• If more than € 100,000 will be collected, the researchers plan to deepen the research on the variable regions of the SARS-CoV-2 genome and other SARS viruses, to propose new drugs in response to any future emergencies caused by other Coronaviruses.

WHAT IS THE CNR-IBF?
Cnr is the largest Italian public research institution, whose multidisciplinarity is represented by a wide variety of institutes. The Institute of Biophysics (https://www.ibf.cnr.it/) studies the structure and mechanisms that make biological systems work. In particular, for years the Structural Biology unit has been working on the search for antiviral drugs, studying their structure and action at the atomic and molecular level. Together with many other internationally known discoveries, about 10 years ago Cnr-Ibf researchers discovered the antiviral properties of a pesticide, which has been recently proved active also against SARS-CoV-2. These skills are essential to target protein structures of viruses and to find drugs on a rational base.

WHAT IS THE ICGEB?
ICGEB is a top-level international research institution with 66 Member States, engaged in innovative research aimed at promoting sustainable development. The Molecular Virology laboratory in Trieste (https://www.icgeb.org/molecular-virology/) studies the mechanisms of infection and develops methods of detecting viruses. This laboratory sequenced and characterized the SARS-CoV-2 genome isolated from one of the first infected Italian patients. Expertise and latest generation techniques are key factors for the study of antivirals on human cells.

Negli ultimi dieci anni, i virus a RNA emergenti continuano a sollevare l’attenzione a livello mondiale: i Coronavirus preoccupano particolarmente, a causa di elevati tassi di mortalità, la mancanza di terapie e la loro abilità ad accendere rapidamente focolai di infezione anche attraverso i confini geografici.

La ricerca di un farmaco efficace contro SARS-CoV-2 e altri Coronavirus emergenti è essenziale per la ripresa in sicurezza delle normali attività e per un futuro immune da una crisi economica mondiale come questa.

Per raggiungere questo obiettivo, è nato il progetto #FarmaCovid, una collaborazione tra i ricercatori dell’Istituto di Biofisica del Consiglio Nazionale delle Ricerche (Cnr-Ibf) a Milano e il Centro Internazionale di Ingegneria Genetica e Biotecnologia (ICGEB) di Trieste.

Ad oggi i risultati ottenuti comprendono:
1 candidato farmaco in test clinici – 1 candidato farmaco in test pre-clinici – diversi composti in fase sperimentale. Non fermate la ricerca ora! La ricerca deve andare avanti.

Il Progetto #FarmaCovid
Grazie ai fondi raccolti e devoluti alle attività del progetto #FarmaCovid, Cnr-Ibf e ICGEB potranno:

• Acquistare un numero crescente di candidati farmaci (15.000 euro) da testare su proteine virali e direttamente su cellule umane infettate dal SARS-CoV-2 (25.000 euro); queste attività sono già iniziate su piccola scala, ma dobbiamo ampliare lo sforzo per aumentare le possibilità di successo e accelerare i risultati di questa prima fase. In base ai fondi raccolti, questa fase potrebbe essere completata entro i prossimi sei mesi.
• Selezionare 2 o 3 farmaci più promettenti, per studiarne attività rispetto alla tossicità (20.000 euro per ciascun farmaco selezionato); questi test sono essenziali per i successivi studi nei pazienti e il loro completamento potrebbe essere raggiunto entro i successivi sei mesi.
• Se si riuscisse a superare la cifra di 100.000 euro, si potrebbe anche approfondire la ricerca sulle regioni variabili nei virus della famiglia SARS, per proporre nuovi farmaci in risposta ad eventuali future emergenze, causate da altri Coronavirus.

Conosci Cnr-Ibf?
Il Cnr è l’istituzione di ricerca pubblica italiana più estesa, la cui multidisciplinarietà è rappresentata da un’ampia varietà di Istituti. L’Istituto di Biofisica (https://www.ibf.cnr.it/) studia la struttura e i meccanismi che fanno funzionare i sistemi biologici. In particolare, da anni l’unità di Biologia Strutturale lavora alla ricerca di farmaci antivirali, studiandone struttura e azione a livello atomico e molecolare. Insieme a tante altre scoperte note a livello internazionale, è di questi ricercatori la scoperta avvenuta circa 10 anni fa di un antiparassitario come possibile antivirale, dimostratosi attivo anche contro il Coronavirus. Questo tipo di competenze è essenziale per raggiungere obiettivi mirati come le strutture proteiche dei virus e identificare farmaci seguendo un criterio razionale.

Chi è ICGEB?
ICGEB è un’istituzione di ricerca internazionale di altissimo livello che conta 66 Stati membri, impegnati nella ricerca innovativa volta a promuovere lo sviluppo sostenibile. Il laboratorio di Virologia Molecolare di Trieste (https://www.icgeb.org/molecular-virology/) studia i meccanismi dell’infezione e sviluppa metodi di rilevazione dei virus. Risultato importante è stato il sequenziamento del genoma virale isolato tra i primi malati italiani di COVID-19 e la sua caratterizzazione. Lunga esperienza e tecniche di ultima generazione sono punti di forza per lo studio di potenziali agenti antivirali.

Dr. Tasneem Elamin (right) graduated in Molecular and Cellular Biology at the University of Milan, at the Structural Biology Unit of Cnr-Ibf tutored by Prof. M. Bolognesi e la Dr. E. Mastrangelo. Tasneem contributed to the study of flavirirus polymerases and spent part of the thesis experience at the Molecular Virology Lab at ICGEB in Trieste, with Prof. A. Marcello (up, center). In particular, she investigated the molecular targets of the anti-flavivirus compounds identified by Dr. M.Milani and Dr. E. Mastrangelo through virtual and biophysical screening.
La Dr. Tasneem Elamin (a destra) si è laureata in Biologia Cellulare e Molecolare all’Università di Milano, svolgendo la tesi di laurea presso l’unità di Biologia Strutturale sotto la supervisione del Prof. M. Bolognesi e la Dr. E. Mastrangelo (Cnr-Ibf). Tasneem ha contribuito allo studio delle polimerasi di flavivirus e ha svolto parte della sua esperienza di tirocinio presso il Laboratorio di Virologia Molecolare – ICGEB di Trieste, con il Prof. A. Marcello (in alto in centro). In particolare, Tasneem ha studiato i bersagli molecolari di un composto diretto contro i flavivirus identificato da M. Milani ed E. Mastrangelo durante screening virtuali e biofisici.

At the Structural biology labs of Cnr-Ibf in Milan, Dr. E. Mastrangelo is engaged in the expression of viral protein targets. Then, isolated targets undergo biophysical characterization in the absence/presence of putative antiviral compounds, to study target-ligand’s affinity and high-resolution structures.
Presso i laboratori del Cnr-Ibf di Milano, la Dr. Mastrangelo è impegnata nella espressione di proteine bersaglio virali. Una volta isolati, le proteine virali vengono caratterizzate in assenza/presenza di composti candidati antivirali, per studiare l’affinità di tali composti per i bersagli e le strutture proteina/composto ad alta risoluzione.

Dr. M. Milani is virtually screening libraries of compounds for their ability to interact with viral proteins as polymerases. This technique, namely virtual docking, starts from the structure of viral proteins at atomic resolution and allows the selection of potential ligands, to be profiled and tested as antivirals.
Il Dr. M. Milani è impegnato nello screening virtuale di librerie di composti, per valutare la loro abilità di legare proteine virali come la polimerasi. Questa tecnica, detta docking virtuale, parte dalla struttura tridimensionale delle proteine virali a risoluzione atomica e permette di selezionare poteziali ligandi, da testare come antivirali.
