<div>Jan leidet an einem sehr seltenen Gendefekt (TMEM63B)

<div>Diese Krankheit wurde das erste Mal 2023 beschrieben und es sind zum aktuellen Zeitpunkt nur 20 weitere Fälle weltweit bekannt. Zudem gibt es nur wenige Informationen oder Erfahrungen zu diesem Gendefekt.

<div>Der Gendefekt zeichnet sich durch eine schwere Epilepsie, eine stark verkürzte Lebenserwartung, Balkenmangel, erweiterte Liquorräume, Hypotonie, Hyperreflexie, Sehstörung/Blindheit, Blutarmut und eine hohe Anfälligkeit für Infekte aus.

<div>Jan kann seinen Kopf nicht heben/halten oder sich drehen. Krabbeln oder gar Laufen wird er möglicherweise nie lernen und ist leider in Zukunft auf einen Rollstuhl und ständige Pflege angewiesen.

<div>Jan kann nicht selbstständig essen oder gezielt greifen, allgemein sind seine Bewegungen eher unkoordiniert. Der Gendefekt und die Blindheit erschweren den Lernprozess, dennoch oder besser genau deswegen, versuchen wir alles in die Wege zu leiten, was ihn dabei unterstützen könnte.

<div>Da es wenig Erfahrung mit dieser Krankheit gibt, bewegen wir uns da auf Neuland und sind auf „ausprobieren“ angewiesen.

<div>Durch seine Erkrankung darf Jan nicht alleine sein bzw. muss ständig beobachtet werden. Aufgrund seiner Epilepsie mit Apnoe kommt es vor, dass er von jetzt auf gleich aufhört zu atmen, weshalb er mittels Monitor 24/7 überwacht wird. Bereits drei Wiederbelebungen haben wir bisher durchmachen müssen. Leider kann es dabei zu einem Status epilepticus kommen, also Anfälle, bei denen das Notmedikament nicht hilft.

<div>Diese dauern länger als fünf Minuten und sind in Verbindung mit seiner Apnoe mehr als lebensbedrohlich und würden seinen Tod bedeuten. Das sind Situationen, die wir definitiv keinen Eltern wünschen.
</div>

<div>Wofür wir eure Hilfe brauchen:

<div>Zum aktuellen Zeitpunkt, gibt es keine Heilung und auch keine genaue Prognosen zu seiner Entwicklung. Jedoch möchten wir unserem Kleinen alles ermöglichen, was ihm helfen könnte, um ihm so viel Lebensqualität wie möglich zu schenken.

<div>Da Jan sich nicht selbst halten kann und immer getragen werden muss, wird er zukünftig auf einen Rollstuhl angewiesen sein. Hierfür werden wir unter anderem bald ein mit dem Rollstuhl befahrbares Auto benötigen.

<div>Zudem ist unsere Wohnsituation alles andere als barrierefrei.
</div>
<div>Somit sind wir gezwungen, ein neues Zuhause mit ausreichend Platz für Jans Zukunft zu finden.
</div>
<div>Jan braucht ausreichend Platz für seine medizinischen Geräte, sein Pflegebett und vor allem die Möglichkeit, dass er am alltäglichen Familienleben teilhaben kann – trotz seiner Einschränkungen.

<div>Durch die Anschaffung eines neuen Überwachungsgerätes, dass die Herzfrequenz und die Sauerstoffsättigung ohne Kabel messen kann, wären wir mit dem Kleinen wesentlich mobiler.

<div>we are Martina and Daniel, the parents of little Jan (almost  3 years old).
</div>
<div>We urgently need your help!

<div>Jan is seriously ill!
</div>
<div>Jan has a very rare genetic defect (TMEM63B).
</div>
<div>This disease was first described in 2023, and currently, only 20 other cases are known worldwide. Additionally, there is very little information or experience regarding this genetic defect.
</div>

</div>
<div>The genetic defect is characterized by severe epilepsy, a significantly shortened life expectancy, agenesis of the corpus callosum, enlarged cerebrospinal fluid spaces, hypotonia, hyperreflexia, visual impairment/blindness, anemia, and a high susceptibility to infections.
</div>
<div>Jan cannot lift or hold his head or turn it.
</div>
<div>He may never learn to crawl or walk and will unfortunately rely on a wheelchair and constant care in the future. Jan cannot eat independently or grasp objects purposefully; in general, his movements are rather uncoordinated.
</div>
<div>The genetic defect and blindness complicate the learning process; however, or rather precisely because of this, we are trying to initiate everything that could support him.
</div>

<div>Since there is little experience with this disease, we are venturing into uncharted territory and are reliant on "trial and error." Unfortunately, Jan cries a lot, and sometimes we do not know why.
</div>
<div>There was once talk of possible phantom pain, but at this point, no one can tell us for sure. Due to his condition, Jan cannot be left alone and must be constantly monitored.
</div>
<div>Because of his epilepsy with apnea, there are times when he suddenly stops breathing, which is why he is monitored 24/7.
</div>
<div>We have already had to go through three resuscitations. Unfortunately, this can lead to status epilepticus, meaning seizures where the emergency medication does not help.
</div>
<div>These last longer than five minutes and are more than life-threatening in connection with his apnea, which would mean his death. These are situations we definitely do not wish upon any parents.
</div>

<div>At this point, there is no cure and no accurate predictions regarding his development. However, we want to provide our little one with everything that could help him, in order to give him as much quality of life as possible.
</div>

<div>Since Jan cannot hold himself up and must always be carried, he will rely on a wheelchair in the future.
</div>
<div>For this, we will soon need a car that can accommodate a wheelchair.
</div>
<div>Additionally, our living situation is far from wheelchair accessible, and we will need a wheelchair accessible house or apartment in the future.
</div>

<div>By acquiring a new monitoring device that can measure heart rate and oxygen saturation without cables, we would be much more mobile with our little one.
